What is this medication?

DIACOMIT (Stiripentol)

DIACOMIT (Stiripentol) is indicated for the treatment of Dravet Syndrome.

How does this medication work? 

DIACOMIT (Stiripentol) has an unknown mechanism of action.  It is thought to enhance GABAergic inhibitory transmission by modulation of GABA-A receptors.

How should I take this medication? 

Oral: Start at 25-50 mg/kg/day given in 2 or 3 divided doses then increase gradually to a maximum dose of 3 g/day.

How to Take: Administer in 2 or 3 divided doses daily with a meal. Capsule should be swallowed whole with a glass of water. Powder for suspension should be mixed with a glass of water and consumed immediately. After administering suspension, add a small amount of water (eg, 25 mL) to the dosing cup and drink the entire mixture to be sure there is no medication left in the cup.  Systemic exposure is slightly higher with the powder for suspension than that observed with capsules. Powder for suspension contains phenylalanine; use caution in patients with phenylketonuria. Powder for suspension contains phenylalanine; use caution in patients with phenylketonuria.

What should I watch for while using this medication? 

Before starting DIACOMIT (Stiripentol), make sure your physician is aware of any allergies or medications you currently take, if you have hepatic impairment, renal impairment, are pregnant, or breastfeeding. Stiripentol use has been associated with adverse events such as loss of appetite, weight loss, blood dyscrasias, CNS depression, and suicidal ideation.

Significant drug interactions exist, requiring dose/frequency adjustment or avoidance so avoid starting any new medications without consulting prescriber.  Anticonvulsants should not be discontinued abruptly because of the possibility of increasing seizure frequency; therapy should be withdrawn gradually (over at least 1 month) to minimize the potential of increased seizure frequency, unless safety concerns require a more rapid withdrawal.  

What if I miss a dose?

If you miss a dose of medication, try to take it as soon as possible. However, if it is almost time for your next dose, take only that scheduled dose. Do not take double or extra doses.

How should I store this medication?

Keep out of the reach of children at all times. Store at room temperature, 59 to 86° F (15 to 30° C). Protect from light. Keep the container tightly closed. Properly dispose of any unused medication after the expiration date.

What are the possible side effects of using this medication?

Drowsiness, agitation, ataxia, hypotonia, dysarthria, insomnia, weight loss, decreased appetite, nausea, decreased platelet count, neutropenia, tremor, aggressive behavior, fatigue, weight gain, vomiting, sialorrhea, bronchitis, nasopharyngitis, fever.
