What is this medication?

Ketoconazole (Oral)
Antifungal Agent

Indicated for treatment of cutaneous candidiasis, seborrheic dermatitis, tinea corporis, cruris, pedis.

Only use ketoconazole when other antifungal therapy is not readily available or tolerated and the potential benefits outweigh the risks.  Serious hepatotoxicity causing fatality or requiring liver transplantation have occurred with the use of oral ketoconazole.  Avoid using ketoconazole with dofetilide, quinidine, pimozide, cisapride, methadone, disopyramide, dronedarone and ranolazine because of the risk of QT prolongation.

How does this medication work? 

Ketoconazole is an azole antifungal that inhibits the biosynthesis of triglycerides and phospholipids and cytochrome P450 enzyme required for the conversion of lanosterol to ergosterol.  This results in increased fungal cell permeability due to reduced amounts of ergosterol present in the fungal cell membrane.

How should I take this medication? 

Oral: Take 200mg to 600mg daily in 2 to 3 divided doses. 

How to Take: Take 2 hours prior to any medications that may decrease absorption due to high pH of gastric contents.

What should I watch for while using this medication? 

Before starting ketoconazole make sure your physician is aware of any allergies or medications you currently take, existing medical conditions, activities you perform.  Ketoconazole can suppress adrenocortical function, increase bone fragility leading to fracture, increase the risk of myopathy.  Avoid taking ketoconazole concomitantly with drugs (proton pump inhibitors, antacids, H2-blockers) that reduce acidity, and to treat fungal meningitis.

What if I miss a dose?

If you miss a dose of medication, try to take it as soon as possible. However, if it is almost time for your next dose, take only that scheduled dose. Do not take double or extra doses.

How should I store this medication?

Keep out of the reach of children at all times. Store at room temperature, 59 to 86° F (15 to 30° C). Protect from light. Keep the container tightly closed. Properly dispose of any unused medication after the expiration date.

What are the possible side effects of using this medication?

Orthostatic hypotension, peripheral edema, fatigue, insomnia, malaise, nervousness, paresthesia, pruritus, alopecia, dermatitis, erythema, erythema multiforme, skin rash, urticaria, xeroderma, hot flash, hyperlipidemia, menstrual disease, nausea, vomiting, abdominal pain, anorexia, constipation, dysgeusia, dyspepsia, flatulence, increased appetite, tongue discoloration, upper abdominal pain, xerostomia, jaundice, myalgia, weakness.
