What is this medication?

SOMAVERT (Pegvisomant)
Growth Hormone Receptor Antagonist

SOMAVERT (Pegvisomant) is a growth hormone receptor antagonist indicated for the treatment of acromegaly in patients who have had an inadequate response to surgery or radiation therapy, or for whom these therapies are not appropriate.

How does this medication work? 

SOMAVERT (Pegvisomant) is an analogue of human growth hormone, Pegvisomant selectively binds to growth hormone (GH) receptors, blocking the binding of endogenous GH, leading to decreased serum concentrations of insulin-like growth factor-1 (IGF-I) and other GH-responsive proteins.

How should I take this medication? 

SQ: Initial 40mg as a single loading dose, followed by 10 mg once daily beginning the day after the loading dose.

Dosage adjustment: Adjust dose by 5 mg increments or decrements in 4- to 6-week intervals based on insulin-like growth factor 1 concentrations (recommended maintenance range: 10 to 30 mg/day; maximum dose: 30 mg/day).

How to Take: For Subcutaneous (SQ) administration only. To minimize the risk for lipohypertrophy, rotate injection site daily, if 2 injections are required, select a different injection site for second injection. May administer in upper arm, upper thigh, abdomen, or buttocks, do not rub injection site. Do not use on area of skin with rash, lumps, bruising or on broken skin. The manufacturer recommends the initial dose be administered under the supervision of prescribing healthcare provider.

What should I watch for while using this medication? 

When starting SOMAVERT (Pegvisomant), make sure your physician is aware of any allergies or medications you currently take, and prior medical history. Pegvisomant may increase liver function tests and should be used with caution in patients with hepatic impairment. Pegvisomant can cause lipohypertrophy following injection and rotation of the injection site should be done to prevent or reduce the risk of occurrence.

What if I miss a dose?

If you forget to give yourself an injection, skip the missed dose and continue with your regular dosing schedule. Do not inject a double dose to make up for a forgotten injection.

How should I store this medication?

If the room temperature is below 25°C:
All boxes of vials may be stored at room temperature (below 25°C). The vials must be protected from light and should not be placed back into the refrigerator. The SOMAVERT (Pegvisomant) powder vials must be discarded if not used within the 30 days of room temperature storage or the expiry date printed on the carton, whichever is earlier.

If the room temperature is above 25°C:
Store all boxes of vials of SOMAVERT (Pegvisomant) powder in a refrigerator (at 2 to 8 °C). Protect the vials from light. Protect from freezing.

What are the possible side effects of using this medication?

Pain, diarrhea, nausea, abnormal hepatic function tests, antibody development, infection, local injection site reaction, flu-like symptoms, chest pain, hypertension, peripheral edema, and sinusitis.
