What is this medication?

TUKYSA (Tucatinib)
Antineoplastic Agent, Tyrosine Kinase Inhibitor

TUKYSA (Tucatinib) is officially indicated for the treatment of breast cancer, HER2 positive, advanced, unresectable or metastatic.

How does this medication work? 

TUKYSA (Tucatinib) is a tyrosine kinase inhibitor that binds to human epidermal growth factor receptor 2 (HER2) and subsequently inhibits MAPK and AKT signaling and cancer cell proliferation to help stop or slow the spread of cancer.

How should I take this medication? 

Oral: Take 300mg twice daily in combination with trastuzumab and capecitabine until disease progression or until toxicity occurs.

How to Take: Take with or without food, every 12 hours.  Swallow tablets whole; do not crush, chew or split.  Can take Tucatinib at the same time as Capecitabine in the days that Capecitabine is also scheduled.

What should I watch for while using this medication?

Before starting TUKYSA (Tucatinib) make sure your physician is aware of any allergies or medications you currently take, and current medical conditions. Tucatinib has been reported to cause severe diarrhea which can result in dehydration, acute kidney injury, and death.  Severe hepatotoxicity has been reported with using Tucatinib.  Monitor hepatic enzymes (ALT, AST) and bilirubin at baseline and every 3 weeks during treatment.

What if I miss a dose?

If you miss a dose of medication, try to take it as soon as possible.  However, if it is almost time for your next dose, take only that scheduled dose.  Do not take double or extra doses.

How should I store this medication?

Keep out of the reach of children at all times. Store at room temperature, 59 to 86° F (15 to 30° C). Protect from light. Keep the container tightly closed.  Properly dispose of any unused medication after the expiration date.

What are the possible side effects of using this medication? 

The most common side effects are diarrhea, palmar-plantar erythrodysesthesia, nausea, fatigue, hepatotoxicity, vomiting, stomatitis, decreased appetite, abdominal pain, headache, anemia, and rash.
