What is this medication?

VOTRIENT (Pazopanib)
Antineoplastic Agent, Tyrosine Kinase Inhibitor Vascular Endothelial Growth Factor (VEGF) Inhibitor

Severe and fatal hepatotoxicity has been observed in clinical trials. Monitor hepatic function and interrupt, reduce, or discontinue dosing as recommended.

How does this medication work? 

VOTRIENT (Pazopanib) is a tyrosine kinase (multikinase) inhibitor.  It limits tumor growth via inhibition of angiogenesis by inhibiting platelet-derived growth factor receptors, cell surface vascular endothelial growth factor receptors, cytokine receptor, interleukin-2 receptor inducible T-cell kinase, fibroblast growth factor receptor, lymphocyte-specific protein tyrosine kinase, and transmembrane glycoprotein receptor tyrosine kinase.

How should I take this medication? 

Oral: Take 800 mg once daily until disease progression or unacceptable toxicity.

How to Take: Administer on an empty stomach, 1 hour before or 2 hours after a meal. Swallow tablets whole.  If concurrent use of a gastric acid-reducing agent cannot be avoided, consider short-acting antacids in place of proton pump inhibitors and H2-receptor antagonists; separate pazopanib administration from short-acting antacids by several hours.  Maintain adequate nutrition and hydration, unless instructed to restrict fluid intake.

You Need to Avoid:  Avoid grapefruit juice.  Do not crush (rate of absorption may be increased; may affect systemic exposure).

What should I watch for while using this medication?

Before starting VOTRIENT (Pazopanib), make sure your physician is aware of any allergies or medications you currently take, if you are pregnant, or breastfeeding. 
Monitor serum liver tests before initiation of treatment with Pazopanib and at least once 59 every 4 weeks for at least the first 4 months of treatment or as clinically indicated. Periodic 60 monitoring should then continue after this time period.  Pazopanib use has been associated with rare but serious adverse events such as cardiotoxicity, GI perforation/fistula, hand-foot skin reaction, hemorrhage, hepatotoxicity, hypertension, infection, ocular toxicity, posterior reversible encephalopathy syndrome, proteinuria, pulmonary toxicity, QT prolongation, skin depigmentation, thromboembolic events, thrombotic microangiopathy, thyroid disorders, tumor lysis syndrome, and wound healing complications.  Watch for any of the following signs and symptoms suggestive of liver problems to your physician right away: yellowing of the skin or the whites of the eyes (jaundice),  unusual darkening of the urine, unusual tiredness, right upper stomach area pain. 

What if I miss a dose?

If you miss a dose of medication, try to take it as soon as possible. However, if it is almost time for your next dose, take only that scheduled dose. Do not take double or extra doses.

How should I store this medication?

Keep out of the reach of children at all times. Store at room temperature, 59 to 86° F (15 to 30° C). Protect from light. Keep the container tightly closed.  Properly dispose of any unused medication after the expiration date.

What are the possible side effects of using this medication? 

Bradycardia, cardiac insufficiency, hypertension, peripheral edema, alopecia, exfoliative dermatitis, hair discoloration, hypopigmentation, palmar-plantar erythrodysesthesia, decreased serum albumin, decreased serum glucose, decreased serum magnesium, decreased serum phosphate, decreased serum sodium, increased serum glucose, increased serum potassium, weight loss, abdominal pain, anorexia, decreased appetite, diarrhea, dysgeusia, gastrointestinal pain, nausea, stomatitis, vomiting, hemorrhage, leukopenia, lymphocytopenia, neutropenia, thrombocytopenia, increased serum alanine aminotransferase, increased serum alkaline phosphatase, increased serum aspartate aminotransferase, increased serum bilirubin, dizziness, fatigue, headache, tumor pain, asthenia, musculoskeletal pain, myalgia, cough, dyspnea.
