What is this medication?

ZELBORAF (Vemurafenib)
Antineoplastic Agent, BRAF Kinase Inhibitor

ZELBORAF (Vemurafenib) is a kinase inhibitor indicated for the treatment of patients with unresectable or metastatic melanoma with BRAF V600E mutation as detected by an FDA-approved test.

How does this medication work? 

ZELBORAF (Vemurafenib) is a low molecular weight oral BRAF kinase inhibitor (potent) which inhibits tumor growth in melanomas by inhibiting kinase activity of certain mutated forms of BRAF, including BRAF with V600E mutation, thereby blocking cellular proliferation in melanoma cells with the mutation.

How should I take this medication?

Oral: Take 960 mg every 12 hours; continue until disease progression or unacceptable toxicity.

How to Take: Doses should be administered orally in the morning and evening. May be taken with or without a meal. Swallow whole with a glass of water; do not crush or chew.

What should I watch for while using this medication? 

Before using ZELBORAF (Vemurafenib) make sure your physician is aware of any allergies or medications you currently take, pre-existing medical conditions and activities you perform. Vemurafenib has been reported to cause severe dermatologic reactions including Stevens-Johnson syndrome and toxic epidermal necrolysis. Fibroproliferative diseases, specifically Dupuytren contracture and plantar fascial fibromatosis have been reported with Vemurafenib use. Vemurafenib can cause hepatotoxicity and requires monitoring at baseline, and during monthly therapy. Vemurafenib has been reported to also cause nephrotoxicity, ocular toxicity including uveitis, blurred vision and photophobia. Vemurafenib has been associated with pancreatitis, photosensitivity, QT prolongation and radiation sensitization.

What if I miss a dose?

If you miss a dose of medication, try to take it as soon as possible. However, if it is almost time for your next dose, take only that scheduled dose. If vomiting occurs after a dose is taken, do not take an additional dose; continue with the next scheduled dose. Do not take double or extra doses. 

How should I store this medication?

Keep out of the reach of children at all times. Store at room temperature, 59 to 86° F (15 to 30° C). Protect from light. Keep the container tightly closed. Properly dispose of any unused medication after the expiration date.

What are the possible side effects of using this medication?

Prolonged QT interval on ECG, hypertension, peripheral edema, fatigue, peripheral sensory neuropathy, headache, maculopapular rash, alopecia, skin rash, skin photosensitivity, xeroderma, palmar-plantar erythrodysesthesia, pruritus, nevus, sunburn, papular rash, erythema, diarrhea, nausea, vomiting, decreased appetite, constipation, dysgeusia, cutaneous papilloma, keratoacanthoma, squamous cell carcinoma of skin, increased gamma-glutamyl transferase, arthralgia, myalgia, limb pain, back pain, musculoskeletal pain, weakness, increased serum creatinine, cough, fibrosis, fever.
