What is this medication?

ZOLINZA (Vorinostat)
Antineoplastic Agent, Histone Deacetylase (HDAC) Inhibitor

ZOLINZA (Vorinostat) is indicated for the treatment of cutaneous manifestations in patients with advanced cutaneous T-cell lymphoma (CTCL) who have progressive, persistent or recurrent disease subsequent to prior systemic therapies.

How does this medication work?

ZOLINZA (Vorinostat) inhibits histone deacetylase enzymes. This group of enzymes catalyze acetyl group removal from protein lysine residues. Histone deacetylase inhibition results cell growth termination and apoptosis.

How should I take this medication?

Oral: Take 400 mg once daily until disease progression or unacceptable toxicity

How to Take: Follow instructions provided by physician/pharmacy label, take consistently at same times each day unless otherwise directed, store at room temperature. Administer with food and maintain adequate hydration (≥2 L/day fluids) during treatment.

You Need to Avoid:  Significant drug interactions exist, requiring dose/frequency adjustment or avoidance so avoid starting any new medications without consulting prescriber. Do not open, crush, break, or chew capsules.

What should I watch for while using this medication?

Before starting ZOLINZA (Vorinostat) make sure your physician is aware of any allergies or medications you currently take, if you have hepatic impairment, are pregnant, or breastfeeding. Vorinostat use has been associated thromboembolism. Monitor for signs and symptoms of pulmonary embolism and deep vein thrombosis. Monitor blood counts every 2 weeks during the first 2 months of therapy and monthly thereafter. Other rare but serious adverse events such as bone marrow suppression, dizziness, fatigue, gastrointestinal toxicity, hepatitis B exacerbation, hyperglycemia, and QT prolongation can occur.

What if I miss a dose?

If you miss a dose of medication, try to take it as soon as possible. However, if it is almost time for your next dose, take only that scheduled dose. Do not take double or extra doses.

Keep out of the reach of children at all times. Store at room temperature, 59 to 86° F (15 to 30° C). Protect from light. Keep the container tightly closed. Properly dispose of any unused medication after the expiration date.
